Thus, a differential analysis will be performed in 3D versus 2D cultivation of U-251 MG glial cells after transgenesis of tumor-associated genes CHI3L1 and CHI3L2 before and after long-term treatment with cytotoxic agent (transcription profiles of genes and a number of microRNAs, morphological analysis, migration and proliferative potentials, immunophenotyping). Particular attention will be paid to microRNAs associated not only with oncogenesis but also with inflammation, which may be the primary indicator of the disease, and which can be used as potential markers for early detection of pathology in the blood of patients. We will also determine the change in the expression of endogenous retroviruses (ERVW-1 and ERVFRD-1) and their transcriptional isoforms in 2D and 3D cultures of U251- CHI3L1/CHI3L2 and glioma samples and in cells of the lymphoblastoid line RPMI-8866 under the influence of the transcriptional activator Tat in HIV-1 infection as potential indicators of inflammation and/or carcinogenesis. In addition, we will determine the content of microRNAs and pro- and anti-inflammatory cytokines/chemokines in the serum of patients with COVID-19 associated with inflammation and various parts of cellular immunity.
