Panchuk, Доктор біологічних наук Registration Date 27-10-2025 Organization Institute of Cell Biology of the National Academy of Sciences of Ukraine popup.description1 The project's primary goal is to develop innovative approaches for treatment of drug-resistant metastatic melanoma, which are based on two tumor-specific vulnerabilities of the disease: elevated levels of reactive oxygen species and dependence on aerobic glycolysis (the Warburg effect). To this end, we will combine metabolomic therapies (low-carbohydrate diet and the antidiabetic drug metformin), with pro-oxidant therapies, such as the novel angucycline antibiotic landomycin A. These therapies destroy melanoma cells regardless of the presence of BRAF600 mutations and PD-L1 expression. For accomplishment of these tasks modern molecular biological and biochemical methods will be addressed, such as Western blot analysis, flow cytometry, and determining the metabolic status of cells and oxygen consumption using an Agilent Seahorse Analyzer. The obtained results will be verified in vivo using B16F10 melanoma models and its drug-resistant subline, B16F10/adr. Successful accomplishment of projects’ tasks will allow us to propose novel effective approaches for targeted chemotherapeutic treatment of metastatic melanoma in clinical practice. popup.nrat_date 2025-10-27 Close
