Information × Registration Number 0200U004533, 0196U018115 , R & D reports Title The ecological taxonomy and physiological micromycetes study as creation base of perspective biotechnologies popup.stage_title Head Zhdanova N.M., Registration Date 08-08-2000 Organization Zabolotny Institute of Microbiology and Virology Ukrainian NAS popup.description2 The object of investigations - the micromycetes from different ecological niches (natural and extremal).The aim - creation of the collection of living cultures, the study physiologal-biochemical peculiarities of the fungi-producers of the biological active substrate with special features - oligotrophy, high radioresistance, thermophily; ecological monitoring of radioactive polluted regions and ecological diagnostics of the biodestruction processes of the industrial materials.The department's culture collection has more than 8 000 cultures of 420 species, 12 varieties, of 140 genera. 49 industrial fungal strains for different areas of country farms are supported in active state. The fungal cultures were isolated in the process of monitoring of radionuclides polluted regions of Ukrainian Polesie ( including the 4 block ChNPP,"Shelter" object and radioactive water). Their biological activity ( growth parameters, radioresistance, growth and destroying ability) of the hard substrates was studied. In a rea of theoretical taxonomy using methods of gene taxonomy the investigation of Cladosporium sphaerospermum and Alternaria alternata populations was conducted. There are 3 species-twins from and radiation polluted and unpolluted regions. In area of natural ecology of fungi the Fusarium oxysporum population investigation was conducted that revealed for the first the mechanism of species transition in this group from oligotrophic to parasitic way of life. Protein exchange peculiarity under influence of low intensity gamma - irradiation was investigated.The protein synthesis was activated in the radioactively treated mycelium of alb-mutant of Cladosporium cladosporioides. The rate of 14C leucine incorporation into biomass of investigated strains correlated with positive radiotropism. The dependence between protein synthesis intensity and the availability of melanin protection system in microsystem is assumed. New approaches to ecological diagnostics of biodestruction processes on the level of the high est taxonomic system links as the reflection of micromycetes manyspecies complexes of specific properties were based.The data obtained open new perspective prognosic on the ecomorphes belonging level of some types of micromycetes to biodestructors with definite abilities not only for prevent systems of technical objects working out from corrosion processes, on the contrary for directed search an active agents of difficult accessibility substrates destroying for modern biotechnology needs.
