Aim of the study: To study the mechanisms of neuromediators and neuromodulators involvement in the processes of development of a stress reaction flowing in the presence of modified HPAA function, and search new ways of prevention of injuring effect of the stress under these conditions. Methods of investigation: Biochemical, radioreceptor methods. Results and treir novelty: It has been established that inhibition of HPAA function in the presence of repeated hydrocortisone administration was accompanied by an encrease, in adrenals, of the activity of catecholamine-, hystamine-, adenosine- and GABA- ergic systems, adn by a number of mediator disturbences in the brain. CNC and HPAA reaction to stress in these animals had its own character- differing from that observed in intact rats- of neuromediator pro vision, nomely: the reaction developed in the presence of inhibition of main nevrochemical processes which are responsible for realization of stress-limiting brain function and neuromediator mechanisms on adrenal levels; of a change in the sign of the response of angiotensin system of the adrenohypophysis; of increasing sensitivity of catecholaminergic processes to stress in pituitary and serotininergic processes in the hippocamp. Under these conditions, the hormonal reaction of adrenal cjrtex to stress (swimming) was reduced, in cose of immobilization there was no reaction; and the response of adrenal medulla significantly different from that observed in intact animals. Activation of HPAA function due to hemiadrenalectomy had a considerable influence on hte development of stress-mediator responce of the brain and adrenals. The nature of stressor disturbonces under these conditions differed from that in case of inhibition of HPAA activity both in regional and mediator aspect. In the hypothalamus of h emiaadrenalectom ized rats the response GABA-, adenosin-, and serotoninergic systems was preserved nearly in full in hte absence of any changes in the activity of angiotensin system, and in the presence of an increase in histamine level, wich is not typical of a normal stress reaction. The role of neuromediator mechanisms which are involved in the regulation of functional activity of the hypothalamus by the hippocamp, is modified under conditions of the effect of stress in the presence of HPAA activation. Preventive effects of a number of pharmacological preparations on neuromediators content and metabolism in the brain and adrenals of stressed rats, and on the hormonal reaction to stress in animals with modified HPAA function, differed in many respects from those observed in intact rats. A part of neurochemical disturbances which were observed under the influence of these preparations, may represent those positive ways which would promote normalization of the course of stress reaction in animals under conditions of experimen tal pathology of HPAA.
