The aim of study is a creation on the base on E.coli and cyanobacterial plasmids and also elements of cyanophage LPP-3 DNA vector constructions, may be used for heterologous genes cloning in cyanobacterial cell. The methods of investigatins are: biochemical, molecular-biological, virological. Screening of plasmid of different taxonomical group have been made.Plasmid pSM1 from cyanobacterium Plectonema boryanum is most useful for vector construction. Some properties of the plasmid (molecular lenght, character of restriction endonucleases digest and so on) have been studed. Plasmid pSM1 was cloned in E.coli pBluescript IISK(+) vector and pBSM1 construction was created. With the help of pBSM1 construction pSM1 plasmid has been maped. Subclonong of pSM1 sequence in vector pBR322 gave possibility to create shuttle vector called pSTS1 that can be replicated in both E.coli and P.boryanum cells. Conjugative triparental matings procedure for vector pSTS1 transfer into the cyanobacterial cells with conjugal pRP4 and helper pGJ28 E.coli plasmids was developed.
