PON repeated administration led to irreversible dilation of the studied vascular smooth muscles. Liposomes filled with quercetine (LQ) in dose 0.1 mg/ml (by lipid) promote recovery of vascular smooth muscles tone lost under PON effects. LQ addition in solution for perfusion of the isolated papillar muscle was accompanied with transitory increase in amplitude of isometric contraction with following gradual its inhibition. Rise of PON concentration led to arrhythmia. In rat Langendorfe perfused heart, PON caused a significant lowering in coronal perfusion pressure and left ventricle pressure. It was accompanied with arrhythmia appearing, which persisted even after the preparation washing. In the isolated papillar muscles, verapamile and ruthenium red had no effect on arrhythmia development induced by PON, and tetrodotoxin and nicorandile led to significant time delay or prevention of arrhythmia development. In the meantime, ouabaine strengthened an arrhythmogen effect of PON. LQ added in perfusate before or just after PON administration prevented or delayed disturbances of myocardium contractility and prevented arrhythmia development during reperfusion. PON intravenous injection was accompanied with tachyarrhythmia appearing with a great number of group extrasystoles, decrease in myocardium contractility and lowering in blood pressure. LQ almost completely eliminated these disturbances. Inhibition of heart contractile and pump functions in acute myocardium ischemia and reperfusion, as well as extrasystole development may be caused by PON effects.
