It was shown that H. polymorpha GSH1 gene is homological to Saccharomyces. cerevisiae MET1 gene encoding S-adenosyl-L-methionine uroporphyrinogen III methyltransferase. Transformants with cloned GSH1 gene were obtained and their physiological and biochemical properties were studied. It was shown that Gsh1+ transformants restored growth and ability to synthesize glutathione on GSH-deficient media in the presence of different carbon sources, including methanol, and acquired resistance to cadmium and chromate ions. Unique coiled-coiled structure, which belong to leucine-zipper transcription factors, and motifs responsible for multiprotein complex formation were found in N-terminus sequence of HpGsh1p. It was assumed that Gsh1 protein acts as a transcriptional factor of GSH2 gene encoding the first and limiting enzyme of glutathione biosynthesis, gamma--glutamylcysteine synthetase.
