Aim of study: To evaluate the parameters of in vivo and in vitro dynamics of cytogenetic indices in cancer patients after radiation therapy for establishing the laws of persistence of genome radiation destabilisation in human blood lymphocytes after partial body irradiation. Methods and tools of investigations: cytogenetic analysis of lymphocyte culture, statistical methods of data treatment, microscopes Biolam I, MBI-6. Results and their novelty: At first time the developing of chromosome destabilization was investigated in blood lymphocytes of patients with uterine body cancer during radiotherapy with modificative influence of pre-irradiation tumor criodestruction. At first time the parameters of spontaneous mutagenesis and dynamics of cytogenetic damage after inhomogeneous fractionated irradiation were studied in cell generations in vitro using the modelof long-term culture of patients' lymphocytes. The qualitative and quantitative peculiarities of the initial steps of cytogenetic effects dynamics in vivo were investigated in patients followed up during 6 months after radiotherapy. The restoration of replicative index and the absence of sister chromatid exchanges accumulation in cells of long-term cultures of lymphocytes taken from cancer patients after radiation treatment were shown. The dependence of aberrant cells elimination rate vitro and in vivo on the level of genome injury was estimated. A new method was developed for detecting the radiation-induced genetic instability in human lymphocytes in vivo and evaluating its potential for persisting in subsequent cell generations in vitro; that was based on estimation of frequency of cells with enhanced chromosome fragility.
