It was shown that recombinant proteins expressed in indicated expression systems. The methods of protein purification were developed from lysates of insect cells and lysates of transfected НЕК293 cells. The monoclonal antibodies to N-terminal SH3A domain of Ruk adaptor protein was developed and characterized. The investigations of multiple endogenous Ruk variants expression, its ability to formation of oligomeric complexes and subcellular distribution were performed in cell lines of various tissue origins. It was shown that expression level and localization of Ruk adaptor protein are specific for each of investigated cell lines. The expression profiles of multiple molecular Ruk form were analyzed by monoclonal antibodies in brain tumor of glial origins. The transcripts with sizes 3,5 kb, 2,5kb and 1,5 kb corresponding to Rukl, Rukm та Ruks isoforms were revealed. These isoforms were identified as immunoreactive bands with molecular weights 85, 56 та 16 kDa respectively. Oscillations in the ability of Ruk praline-rich motifs to interact with exogenous ligands were revealed, suggesting the existence of the dynamic equilibrium between active (opened) and inactive (closed, resulted from intramolecular interactions) forms of adaptor protein.
