The object of the study was human lung adenocarcinoma cells of A549 line and human breast adenocarcinoma cells of MCF-7 line, sensitive and resistant to doxorubicin (MCF-7/wt and MCF-7(Dx-R), respectively). Also levels of transcription of antiapoptotic protein Bcl-2 and proapoptotic proteins p53, Bax, Bad under the action of DNA-damaging factors were studied. We found that under the action of X-ray irradiation the level of Bax and Bad expression was increased in MCF-7(wt) cells, though in MCF-7/Dx-R cells it changed insignificantly. It has been demonstrated, that in cells of MCF-7/DOX line another form of Bcl-2 protein was expressed, which differs from the same form in MCF-7(wt) line. We found, that MCF-7/DOX cells are more resistant to growth-inhibitory effect of irradiation, as well as have increased ability to reparate DNA, and the level of mutations in DNA, induced by radiation, was significantly smaller. After irradiation with doses of 3,0 and 4,5 Gr the level of expression of p53 protein was increased in MCF-7/wt cells, but not in MCF-7/DOX cell line. The received results indicate about possible existence of cross-resistance of malignant cells to action of anti-cancer agents and X-irradiation, and also about potential role of TGF-beta regulatory system in action of this cytotoxic agents.
