A technological regulation for obtaining of dendritic cells (DC) from peripheral blood, leukoconcentrate of peripheral blood and wound exudates of cancer patients has been developed. The proper terms for obtaining of DC with taking into account the terms of conducting of polychemotherapy (PCT) in patients with advanced ovarian cancer have been determined. Using tumor antigens (TA) of different nature (lysate of tumor cells (LTC), mechamomodified lyophilized tumor cells (m/m LPTC), synthetic glycoproteins, peptides HER-2/neu) DC-based vaccine has been created. With phenotyping, functional tests and cytomorphological examinations it has been proved that degree of DC maturation depends on the type, method of preparation and concentration of TA used for their loading. Anti-tumor and antimetastatic activity of DC dependently on the way of TA "loading" and ways of administration in experiment has been examined. Intravenous (i/v) infusion of DC "loaded" with LTC to mice with sarcoma 37 and intracutaneous or i/v administration of DC "loaded" with LPTC or m/m LPTC to mice with Lewis' carcinoma are shown to be the most effective. Increase of the amount of infusions of DC-vaccines promotes its antimetastatic effect. Realization of anti-tumor and antimetastatic effect of DC-vaccines occurs at the expense of stimulation of lymphoid tissue of the recipient. Increase of lymphoblasts and total amount of cells in the thymus, immunoblasts, plasmatic cells, CD3+, CD4+,CD8+ - lymphocytes in the spleen, total amount of lymphocytes, CD8+ - lymphocytes and BGL in peripheral blood as well as significant increase of cytotoxic activity in splenocytes take place. Decrease of antimetastatic effectivity of DC- vaccines is accompanied by the increase of the ratio CD4+/CD8+-lymphocytes in the spleen and decrease of functional activity of DC in vitroin the reaction of mixed culture of leucocytes with alogenic lymphocytes.
