Methods of cell cultivation, cytogenetical and statistical methods have been used in the work. It has been shown the karyotypic heterogeneity of cells of the investigated cell lines and its diminution during long-term cell in vitro cultivation, chromosomal instability, including structure chromosome rearrangements and chromosome number changes, and increasing of chromosomal instability during stem cells in vitro cultivation. It has been shown that cell lines of mouse embryonic germ cells accumulated genetical changes during in vitro cultivation, and adapted for conditions of cultivation. The cell adaptation was accompained by spontaneous neoplastic cell transformation and intensive karyotypic evolution of investigated cells in culture. The further study of the correlation between the mitotic checkpoint loss, the special features of karyotype changes and neoplastic cell transformation has the important value for understanding the mechanisms of chromosomal instability, which is a common hallmark of transformated cells.
