To establish the specificity of domain organization, we studied alpha-globin domain in leukemic cells and cells of other lineage. In the course of the project realisation, new methodical approaches were developed, genomic oligonucleotide DNA microarray in particular. Such microarray consists of an arrangement of unique DNA fragments representing the whole domain of chicken alpha-globin genes that allow studying dynamic changes in chromatin domains organization and functioning. We determined the transcriptional status of alpha-globin genes domain and mapped the attachment sites with nuclear matrix. Differences in spatial organization of alpha-globin domain in leukemic and lymphoid cells were found in particular domain in leukemic tissues was only a half in length comparing to the normal. Initialtranscription zones and dynamics were identified within the chromatin domain. Chromatin domain structure and function elucidation during differentiation and tumorogenesis revealed changes in domains size, specificity of nuclear matrix attachment, and transcriptional status.
