Information × Registration Number 0207U005432, 0103U001714 , R & D reports Title Biotechnological production and functional characteristics of proteins important for diagnostics and medicine popup.stage_title Head Sibirny Andrey Andrejovych, Registration Date 10-04-2007 Organization Institute of cell biology, National Academy of Sciences of Ukraine popup.description2 The investigations on the project were carried out in 4 main directions: 1) Development of producers of heterologous and homologous proteins of bioanalytical and medical significance using the methylotrophic yeasts (the aim: a) to conduct a selection of gene-engineered strains of Aspergillus niger producers of glucoseoxidase, and Hansenula polymorpha producers of human pro-insulin, surface antigen of hepatitis B, aminooxidase, and gluthation; b) to develop effective systems for multi-copy gene integration for obtaining over-producers of heterologous proteins of diagnostic and treatment importance); 2) Biotechnologic production and structure-enzymatic characteristics of enzymes of diagnostic significance (the aim: a) to select yeast strains overproducing oxidoreductases of diagnostic significance; b) to study regulation of enzymes' biosynthesis, and develop effective systems of their purification; c) to isolate highly purified oxidoreductases, characterize them and develop novel methods for clinical diagnostics based on these enzymes); 3) Study of the ways for increasing effectiveness of phagocytosis performed by macrophages and neutrophilic blood cells (the aim: a) to optimize conditions of performing phagocytosis of polystyrol nanoparticles by macrophages; b) to study a possibility of modulating phagocytic activity of cells by the biologically active substances of natural origin); 4) Search for new tumor-associated antigens and tumor-specific markers using adapter protein Ruk/CIN85 as an experimental model (the aim: a) to purify preparative quantities of recombinant adapter proteins and develop monoclonal antibodies against studied antigens; b) to study an expression of Ruk/CIN85 in samples of human prostate at the level of mRNA and protein). The research tasks on the indicated items of the aim have been fulfilled. 47 articles in Ukrainian and international scientific journals were published taking into account the results of the project.
