The purpose of the project is the elaboration metods of genetic and gen-engineering construct which produces anticancer antibiotic nogalamycin Streptomyces nogalater and obtains strains with enhanced product of this drug. Metods of investigations - determination antibiotic activity with test-culture, study of the resistance strains to antibiotics with disks, editing vwith mutagens, discharge of total and plasmid DNA, PCR, E. coli - Streptomyces conjugation, DNA-DNA hybridization. The conditions for thin-layer chromatography construction are determinate. The collection of antibiotico-resistant mutants S.nogalater DSM40546 і S.nogalater SU26 is obtained and studied. The frequency of conjugational transference from cells of E. coli to cells of S. nogalater replicate and integrate plasmids is investigated.The fragments of claster biosynthesis nogalamycin are amplificated.
