Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 The panel of human solid tumor samples paired with control samples of non-transformed tissues as well as blood serum samples of the oncologic patients has been collected. The screening of tumor samples of different localization (including lung, ovary, mammary gland, endometrium, cervix, vulvae, bladder, stomach, esophagus, thymus) for human beta-defensin-2 (hBD-2) mRNA expression was performed. It has been shown that in the majority of tumors (excluding tumors of thymus, mammary gland, ovary, bladder) hBD-2 mRNA is overexpressed compared with paired control samples. The regimen of recombinant hBD-2 production and purification with the use of affinity and reverse-phase chromatography was developed. Using rec-hBD-2 as antigen, immunization of animals was carried out and monospecific polyclonal anti- hBD-2 antibodies and anti-hBD-2-mAbs were generated. Different variants of immunoenzyme assay (ELISA) utilizing monoclonal and polyclonal antibodies were developed and tested against standards of rec-hBD-2 dilutions. Analysis of clinical materials (blood serum of oncological patients) by ELISA has demonstrated that hBD-2 levels in blood serum don't correlate with its expression level in tumor samples. The developed technique may be used for evaluation of hBD-2 content in biologic solutions (cell and tissue lyzates, conditioned media).
