PKD2 differential expression in subpopulations of mature human B lymphocytes was shown for the first time. Memory B cells and plasmablasts were characterized by highest level of PKD2 expression, while plasma cells demonstrated the lowest level of PKD2 expression. The peak of PKD2 activity was revealed in germinal center B cells. The kinetics of PKD2 activation via B cell receptor, CD40 and CD150 was clarified. For the first time our studies showed that BCR engagement resulted in rapid PKD2 autophosphorylation and relocalization to the cell nucleus. Also we found nuclear localization of autophosphorylated PKD2 in activated normal and malignant B lymphocytes in situ. Using immunohistochemical approach we found that diffuse large B cell lymphomas could be subdivided into distinct groups based upon the level and intracellular localization of autophosphorylated PKD2.
