Tarady Nelli, Registration Date 02-04-2009 Organization International Center Astronomical, Medical and Ecological Research popup.description2 The expression, dislocation, colocalization of proapoptosis (Fas/ CD95+, Bax, intracellular receptors to TNF and IFN ) and antiapoptosis (Bcl-2, IL-2R, iNOs) markers in CD4, CD7, CD8, CD16, CD20, CD57, CD22, CD25 ICC and kidney tissues have been investigated. ICC apoptosis was estimated with the help of annexin V, marked FITC, and PI. Differentiation markers and the apoptosis markers were revealed using monoclonal serums marked by fluorescent labels in green (FITC), yellow (PE) and red (Cy5, PrCP) areas of a spectrum, the cell's nucleuses were painted in dark blue colour by Hoechst or red colour by PI. The results were took into account by multimedia luminescent (Carl Zeizz-250SF, computer program Multichenel AxioVision 4.4) and confocal laser scanning microscopy (Axioskop-2 LSM 5 PASCAL Carl ZEISS with helium-neon and argon lasers) at nongraded three and four-colour colouring. The received images scanned and were processed with the aid of the computer program LSV510. Research was carried out at healthy contingents a of various age groups: children in the age of from 7-14 years (17) and women in the age of from 18-22 years living on plain (21) and in highland (15); and at chronic inflectional inflammatory diseases of kidneys at children (57) with chronic glomerulonephritis GN (38) and pyelonephritis PN (19); at chronic inflectional inflammatory gynaecological diseases at the women, living on plain (Kiev -15, Nalchik 25) and in highland (27). The qualitative and quantitative changes, colocalization, dislocation expressing markers of apoptosis in ICC are established. At PN children chronization of inflammatory process is connected to activization of the expression of iNOs, Bcl2 and decrease of Bax in CD8 on a background of a normal level of expression of intracellular IFN and TNF , and also Fas- marker, effector mechanisms of the apoptosis are more activated in CD4. At GN the expression of markers TNF , INF , iNOs, Bax, Bcl2, FasApo1 and apoptosis in CD4 and CD8 are significant labilized. Before treatmentthe close authentic correlation relationship were established only between the apoptosis of ICC and the expression of Bcl2 in CD4 (r = 0,88, р < 0,02), in CD8 (r=0,55, p < 0,05). After treatment between all markers and the apoptosis the close authentic direct correlation connections from r=0,67 to r=1 (p < 0,016) were established.
