Goal of the study: determing the role of the suppressed melatonin-producting function in the pineal gland in a process of intensive vascular system aging and detecting the probability for the improvement of its revealed functional changes due to natural light regime renewal and/or replacement therapy with melatonin. Methods of the study: physiological, biochemical, radionuclide, immunofermental, histological, morphometrical, clinical, statistical. The study has been carried out either on young matured and old rabbits and rats or old patients. The dynamics of the influence of both age hypopinealism in the old and hypopinealism induced by long-term diurnal illumination in the young on the cardio-vascular system (arterial pressure, electrocardiogram, eye-microvessels) as well as some endocrine mechanisms of its regulation (pineal melatonin-stimulating function HTS and SAS), on chronostructural indices of hormonal HTS disorders and lipid blood spectrum under above - stated light regime renewal, and/or the therapy with melatonin has been studied. In hypopinealism affected patients and animals pathological vascular changes (arterial hypertension, atherosclerosis, chronic cardiac insufficiency, sinus bradycardia), destructive processes in eye microvessels with haemorrhages and necrotic zones formation have been observed against the background of SAS activation. The signs of hypothyroidism and stable desynchronism are more indicative following the extra cholesterol load and can not be normalized by natural light regime renewal. In course administerering melatonin was shown to normalize an increased blood pressure, to improve profoundly eye microstructure, lipid chronoalgoritm and HTS indices under desynchronism. The melatonin monotherapy can be combined with the other antihypertensive medicines (lizinopril, amlodipin). It is advisable to apply to the old hypertensive patients with a dependence upon the stage and severity of the disease caused by the age pineal hypofunction.
