Positive and negative regulators of glutathione biosynthesis have been studied out. Selection method of recombinant strains posessing increased intracellular glutathione level has been developed. Mechanisms of glutathione transport and degradation have been studied. Application possibilities of obtained results for the purpose of hyperproducers construction have been evaluated. H. polymorpha recombinant strains carrying additional copies of glutathione biosynthesis genes have been constructed. Phenotype and glutathione production of these strains have been studied. Effect of various compounds on expression of HpGSH2 gene, encoding enzyme of the first step of glutathione biosynthesis, has been studied. Conclusions of positive/negative regulation of the gene expression have been done. HpGSH2 gene has been modified by means of point mutagenesis and recombinant strains carrying modified gamma-glutamyl cysteinyl synthetase have been constructed. Effect of mutation on glutathione production have been studied.
