Kavetsky Institute of Experimental Pathology, Oncology and Radiobiology popup.description2 Chemically pure aconitine, aconitine-containing plant extract of Aconitum spec.; to carry out the preclinical research of the angiogenesis-mediated anticancer activity of aconitine-containing agents (ACA); methods of experimental oncology, cell culture, biochemical, biophysical, and statistical methods. It is shown, that AVA in low not cytotoxic concentration shows proapoptotic action towards actively proliferating endothelial cells and causes inversion of their surface charge. In this case agents investigated prove as direct inhibitors of tumor angiogenesis. It is revealed, that AVA in high (not cytotoxic in relation to tumor cells) doses, develop anticancer action by anticvascular mechanism. At low concentration (subcytotoxic in relation to active proliferating endotheliocytes) AVA are capable to show direct antiangiogenic anticancer action. Preclinical research of specific pharmacological activity AVA on 4 experimental tumor strains has revealed their ability to inhibit growth and metastasis of solid tumors. Anticancer and antimetastatic effect is dose-dependent and realizes by two mechanisms: antivascular (at total doses close to MTD/2) and antiangiogenic (at total doses more low MTD/30). Antiangiogenic action of AVA provides anticancer and antimetastatic (> 80-90 %) action only concerning tumors with angiogenesis-dependent growth. In accordance with requirements of the National Pharmacological Center of Ukraine concerning carrying out preclinical researches of specific pharmacological activity of anticancer agents it has been shown, that AVA are characterized the significant angiogenesis-mediated anticancer action to tumors with angiogenesis-dependent growth and can be considered as perspective anticancer agent with antiangiogenic action.
