Also the level of resistance of this strain against different antibiotics was tested by a disk diffusion test. Construction of the reporter system for detection of the thiostreptone-like producer strains was performed. The highest level of antibiotic production was achieved in the liquid mediums with saccharose and asparagine or NaNO3. High levels of glucose, nitrogen and phosphate repress siomysin production. Sulfur has a slight influence on antibiotic production. Production of antibiotic starts in the mid-exponential phase and occurs in the growth phase-dependent manner. After 6-7 days production of siomycin severely decreases. We selected effective mutagen dosage that allows to get S. sioyaensis strains with enhanced siomycin production. Selection of the S. sioyaensis strains resistant to different concentrations of aminoglycoside antibiotics and rifampicin is a good approach to get overproducers. We got a lot of strains with increased siomycin production. Various replicative plasmids (pKC1139, pKC1218E, pSOK101) as well as integrative vectors pSET152 and pSOK804, respectively were conjugally transferred from E. coli into S. sioyaensis. Obtained recombinant strains are stable. Presence of these vectors in S. sioyaensis do not influence siomycin production and morphogenesis. The utility of temperature sensitive replicon of pKC1139 for insertional gene inactivation in S. sioyaensis has been validated by disruption of the putative nonribosomal peptide synthetase gene.
