Information × Registration Number 0210U003521, 0108U004128 , R & D reports Title NO-dependent mechanisms of the regulation of morphofunctional state of blood immunecompetent cells under the influence of ionizing radiation. popup.stage_title Head Sybirna Nataliya Oleksandrivna, Registration Date 09-02-2010 Organization Ivan Franko National University of Lviv popup.description2 The object of the investigation - molecular and cellular mechanisms, which designate morphological and functional changes in the peripheral blood of mononuclear and polimorphonuclear leukocytes under the ionizing irradiation, (i.e. degradation of nuclear DNA, changes in the lectin-induced aggregation of neutrophilic granulocytes, proliferative activity of the human peripheral blood leukocytes in the reaction of blast transformation), and possibility to correct pathological changes in the metabolic processes of experimental cells by using direct modeling of the process NO synthesis. It is determined the total activity of NO-synthase and content of NO- metabolites (nitrates and nitrites) in the rat peripheral blood leukocytes both under the low level x-ray radiation intensity (1 cGy/day), and per os administration of the main substrate and non-selective inhibitor of NO-synthase on 10th, 20th, and 30th days of the experiment. It is estimated the impact L-arginine/NO system on the specificity of apoptosis process in the normal state of rat leukocytes and after low level x-ray radiation, by the analyzing the content of fragmented DNA and the comet types. It is determined the increase in p53 protein content and decreases of antiapoptotic protein Bcl-2 during the irradiation by using immunocytochemical methods. In the in vitro experiments of analysis prolipherative activity of human peripheral blood immunocompetent cell, was found that preincubation of immunocopmpetent cell with L-NAME and influence of x-ray irradiation in dosage of 0,3 та 2,0 Gy leads to decrease in reaction of blast transformation. At the same time the preincubation of peripheral blood immunocompetent cell with L-Arg leads to the increase both the apoptosis index and proliferative activity.
