Recombinant strains which contain FMN1 gene (encoding the riboflavin kinase) driven by the strong constitutive promoter TEF1 able to overproduce flavin mononucleotide (FMN) were isolated. Using mutagenesis and selective media the stable strain that has deregulated riboflavin biosynthesis pathway and high level of riboflavin production was selected. Transformation of this strain by the plasmid containing RIB1 and FMN1 genes under the TEF1 promoter resulted in selection of recombinant strains able to produce up to 80-100 mg FMN/L. The methods of statistical experimental designs were used to determine the optimum conditions for FMN production. The strains with high riboflavin kinase activity were obtained as a result of this work. Such strains can be used as a source of this enzyme and are interesting for analytical goals, specifically for determination of ATP. Such approaches (the construction of FMN overproducers by means of substitution of native promoter of FMN1 gene by other strong promoter TEF1), as well as the isolated strains - overproducers of FMN, can be used for construction of other flavin nucleotide - flavin adeninedinucleotide.
