In single intraperitoneal administration LD50 of amitosinoberamide is 300 mg/kg in rats, 148 mg/kg in mice, and 130 mg/kg for hamsters. The acute toxicity of amitosinoberamide is manifested in sympoms characreristic of acute poisonong with natural and modified berberibe compounds. The effects of amitosinoberamide in normal tissues is different from that of alkylating agents. When chronic toxicity was assessed, LD50 of amitosinoberamide in rats amounted to 90 mg/kg suggecting the cumulative effects of the substance. In general, amitosinoberamide is less toxic than berberine and thyophosphamide while its efficacy, in particular, in human malignant lymphoid cells is not inferior. The mechanisms of pharmacologic activity of amitosinoberamide are predominantly associated with the alkaloid moiety of the molecule. Amitosinoberamide is devoid of mutagenic activity in Ames test. Amitosinoberamide is not teratogenic in Wistar rats.
