Object of research - seedlings and seeds of different varieties of winter and spring rape. Purpose of research - the development and introduction of new highly effective methods of selection of cold- and frost-resistant lines and varieties of crucifers; the evaluation of resistance of crucifers genotypes to hypothermia. During the research period (2001-2005) the work on approbation and modification of methods of diagnostics for resistance of crucifers genotypes to effects of hypothermia was conducted; the comparative characteristic based on 4 years results was given and the implementations in selective and production activity of the institute were made. A new method of identification of genetically determined frost resistance in winter crucifers plants was developed on seedlings in two modifications. The modification of the plasmolytic method of determining the degree of plant cold-resistance was conducted, as well as modification of the method for evaluating the frost-resistance of plant varieties by germinating its seeds on solutions with high osmotic pressure and two modifications of the method of cold seed germination. In order to evaluate the collection and selection material of Ivano-Frankivsk Institute of Agroindustrial Production on the resistance of variety samples to the effects of hypothermia four of the developed methods were implemented in production. They were used for analyzing 340 varieties of crucifers, which helped determine the variety groups which are recommended for introducing into the selection process for enhancing cold-resistance. The diagnosis of winter crops was conducted by method of direct calculation of plants that successfully wintered in the field. During the report period 300 variety specimens of crucifers of different origin were sowed out and analyzed. The analyzed material turned out to be very diverse cold- and frost-resistance wise. Based on every studied index the variety groups with different levels of resistance to unfavorable factors were distinguished. Thus the existence of genetically determined differences in resistance of winter rapeseed varieties to the effects of hypothermia is confirmed. The comparative study on methods of diagnosing the cold- and frost-resistance of crucifers was conducted. All of the suggested methods of determining genotype resistance to hypothermia are simple and quick to execute and do not require expensive equipment or depend on season. In order to better compare the results of analysis of material by different methods the diagnostic scale was unified for all of them: five point scale system and five groups of resistance were created. The comparison of results of collection material evaluation conducted by different methods has shown the coincidence of resistance groups in majority (83%) of varieties. The received results of laboratory analysis correlate with winter-resistance of varieties, which was determined under field conditions. The results coincided in 77% of cases. Thus the modified methods of low temperature resistance evaluation for crucifers give well differentiated and coinciding results and they could be recommended for initial mass evaluation of selection material under laboratory conditions. The considerable variability of crucifers genotypes by the reaction to the effect of hypothermia (for cold-resistance - 30%, winter-resistance - 25%, frost-resistance - 55% after congealing and 61% in case of sprouting on osmotically active solution) gives a wide choice range for a breeder in diversification of crossing schemes. These methods are applicable for both individual and variety diagnosis. This diagnosis allows the simultaneous selection of plants that are the most resistant to stress, which is very valuable.
