The object of the research is phytobiota of the south-east of Ukraine as a material system of plants' populations and elements of the system of different organization levels. The aim of the paper is to develop floristic, phytocoenotic and population basis of monitoring, conservation and use of flora, especially in conditions of high anthropogenic pressure. The methods of research are floristic, geobotanical, critical-systematic, morphological, introductive, phytosozological, comparative, historical and gradient analyses. The scheme of phytobiota monitoring in the south-east of Ukraine has been designed, the data base of the region's phytodiversity has been made, which includes 2070 species of natural flora vascular plants. The classification system of natural vegetation of the south-east of Ukraine (10 kinds, 30 formation classes, 539 formations, 2751 aggregations) has been developed. Four plant species, new for the region, and new location of 67 rare species have been found. The herbarium fund consists of 115,316 herbarium pages, carpological collection includes 54.5 thousand samples. The critical-taxonomic revision of the flora composition in the south-east of Ukraine have been published, a new system of Artemisia L. genus within the Ukrainian flora has been introduced. The Red Book of Donetsk Region: Flora has been first published. According to the results of inventory studies of vegetation cover the area of nature-reserve fund has been extended and new objects of nature conservation have been founded. The general structure of the regional ecological network of Donetsk region has been developed. The measures have been taken to retain and improve the "Steppes of Ukraine" exposition, a national heritage of Ukraine, containing 424 species, including 96 rare and disappearing species. It has been proved that burning resulted in increasing steadiness of introducing populations of steppants. The reintroduction method has been proved efficient for conserving rare species of plants in anthropogenic transformed conditions, biological basis of forming reintroduction populations of rare and disappearing plant species has been scientifically grounded.
