Karazin popup.description2 The purpose of this project is the characteristic of the potential oncogenes expression in tumours of a brain as at level RNA, and proteins, and also changes ionic homeostasis in tumoral cells of carcinomas. For performance of the project independent methods - Northern-hybridization, a return transcription - polymerase chain reaction (PCR), immunological the analysis is used. The received results will help to define a role of researched proteins in normal and tumoral cells, their participation in signaling, that practical application for diagnostics and can find of classification of tumours. At the previous stages the raised level of the expression of potential oncogenes HC gp-39, YKL-39 and IGF-II A in tumours of a brain at level RNA, and also raised production of proteins HC gp-39 and YKL-39 in glial tumours of a brain was investigated. Results testify to activation genes HC gp-39 and YKL-39of the expression not only on equal RNA, but also at a level of protein in these tumours. Increased expression of genes YKL-39 and HC gp-39 correlates with the greater degree of malignant of the glial and has potential for use as a molecular marker for prognostic estimations of astrocytic tumours. Received by us recombinant protein GST-HC gp-39 was used for search in eukaryotic cells (a liver of mice) albuminous partners potential oncogene HC gp-39 with the help pull-down assay. As a result of the experiments repeated three times proteins in the size 28 kDa that is potential partner HC gp-39 were revealed.
