There are two 98% similar isoforms of eEF1A (A1 and A2) which are tissue and development-specific. A2, rather than A1, reveals oncogenic properties when appears in non-specific tissue (ovary, pancreas etc.). Why just one of the two 98% similar eEF1A isoforms reveals oncogenic properties remains unknown. Our aim is to unveil the molecular properties of eEF1A2 which could provide a molecular background for its cancer-specific action. We have shown different ability of the A1 and A2 isoforms to interact with SH2 and/or SH3 domains of different signaling molecules as well as with calmodulin which suggests a possibility of non-similar involvement of A1 and A2 in cell regulation. Striking conformational dissimilarity of the isoforms was found by X-ray scattering, сircular dichroism and confirmed by scanning microcalorimetry.
