In research work comparative analysis of basic methods for DNA extraction from bone tissue and its amplification was conducted. The methodological aspects of performing of forensic-medical molecular-genetic research of the object, in which bone tissue of a different structural type and degree of its saving were developed. A number of modifications of basic stages of the genomic DNA extraction from bone tissue of changed (body), exhumed, sceletonized corpses and corpses from fireplace were proposed. The principles and terms of PCR-typing of DNA extracted from bone tissue have been developed. Most optimal bone objects for conducting the identification research were chosen. Methodical recommendations for selection, preparation and terms of storage of bone biological material are offered in work. Practical recommendations for conducting of molecular-genetic forensic-medicine identification examination after bone remains were composed. Modified protocols of the DNA extraction from bone tissue and it PCR-typing are successfully applied in practice of forensic-medicine examination in different regions of Ukraine.
