The aim of investigation was the induction of nerve stem cells direct differentiation to the cholinergic neurons. The methods of cell culture, immunocitochemistry and PCR was used. During experiment nerve cells of human embryo 9-10 weeks of gestation was tested for capacity to differentiate into cholinergic neurons. Using different neuroinductors and their combination we determined, that combination of acethilcholine and retinoic acid was most effective of them. We received 21% of ChAT-positive cells in the culture population. These cells expressed specific genes of cholinogenesis - VAChT and AChE and demonstrated the high level of AChE activity. The obtained data open new perspective for the cell therapy of neurological disorders. So, suspension of nerve cells, enriched by cholinergic neurons and their precursors can be received in culture and using as a material of neurotransplantation.
