For main commercial germplasms (heterotic groups) inbreds with high ability to morphogenic callusogenesis of different types were determinated which can serve the basic in their subgermplasms and germplasms for experiments on optimization of regeneration process, somaclone obtaining, purposeful genetic transformation of specific heterotic groups. Profound investigations of effects of such abiotic factors as low positive temperatures, high concentrations of sodium chloride, herbicide dicumba, aminoacids on callus tissue cultures in regeneration period and in embryoculture were carried out. The main methodic approaches to the creation of genotypes in in vitro culture tolerant to stress abiotic factors were elaborated. According to theoretical and practical developments biotechnologies of obtaining callus tissue of types I and II and plant regeneration were created, selective system for identification of genotypes tolerant to low positive temperatures on the ground of embryoculture was carried out, the biotechnology of preservation of elite and initial breeding materials in cold conditions was suggested, the method of obtaining cell cultures by direct and indirect techniques was improved. Obtained regenerated plants were transferred to breeding divisions as initial material. Induced callus cultures were used for search works on genetical transformation of maize lines of Ukrainian selection.
