Voloshchuk, Registration Date 14-02-2011 Organization The V.M. Remeslo Myronivka Institute of Wheat popup.description2 Object: regenerated plants and callus tissue of winter wheat, fungal pathogen isolates. Purpose: to develop methods for induction of somaclonal variants of systemic acquired resistance (SAR) genes. Methods: tissue culture, cell selection, methods of biochemistry, biochemical genetics, microbiology and mycology. Results. Differentiation in activity level of peroxidase and changes in conductivity between resistant and susceptible winter wheat genotypes when infecting regenerated plants with necrotrophic fungal pathogens has been revealed. Treating ears with antioxidants and addition of antioxidants to nutrient media has been proved to increase callus induction in wheat anther culture. The somaclonal variants with increased expression of genes associated with SAR have been obtained. The relation between resistance to fungal pathogens and accumulation of phenolic compounds and polyphenoloxidase and phenilalanilammonialiase activity has been found. These somaclonal lines being tolerant to pathogens may be mutants for SAR genes. Field of use: wheat breeding.
