The aim of the project: to study tertiary structure of octameric flavin-dependent enzyme alcoholoxidase from the methylotrophic yeasts Hansenula polymorpha using X-ray crystallography to obtain a new information about relations between oligomeric protein structure and its catalytic function The methods of investigation: microbiological, biochemical, physical, physico-chemical, enzymological, analytical. It was developed the technology of isolation and purification of alcohol oxidase from the over-producing strain of the methylotrophic yeast Hansenula polymorpha C-105 (gcr1 catX), as well as from the mutant H. polymorpha strains with decreased affinity of alcohol oxidase to ethanol. Some physico-chemical and enzymological properties of alcohol oxidase preparations purified from the studied strains were estimated. Highly-purified alcohol oxidase preparations were transferred to Russian partner (A.N. Bach Institute of Biochemistry, Russian Academy of Sciences, Moscow) for enzyme crystallization and investigation of tertiary structure using X-ray crystalography. The analysis of difraction data for alcohol oxidase was performed together with Russian partner. The modeling of potential changes in protein structure, important for site-directed modification of H. polymorpha AOX gene to obtain the modified alcohol oxidase forms with essential decreased affinity to ethanol was done.
