Danilevsky Institute of Endocrine Pathology Problems of Academy of Medical Sciences of Ukraine" popup.description2 The object of the investigation is thyroid status of rats with experimental merkazolil hypothyroidism and Chinchilla rabbits with experimental AIT and hypothyroidism after the influence of biologic preparations of placenta and chemical compound YAV-99; synthesis and specific affect of compounds with thyrotropic activity. The purpose of the investigation is: foundation of the of cryoconserved biological placental preparations use for the correction of the thyroid pathology and restoration of the thyroid function in an experiment; the identification of specific features of thyroid-stimulating activity of synthetic analogues of natural compounds. Methods of theinvestigation: pharmacological, immunsorbent, biochemical, chemical, physical-chemical, biological, histological, statistical. It is shown that placentas alo- and ksenotransplantation leads to the normalization of the hipophysal and thyroid systems state of rabbits with experimental hypothyroidism. The affect of alo-preparation was more demonstrative and prolonged. In both cases on the remote terms of research the positive changes of thyroidal system and thyroid histological structure are marked. The efficiency of chemical compound YAV-99 within monthly introduction on the rabbits' thyroid function with an experimental hypothyroidism proceeding is high enough. In rabbits with experimental AIT placenta transplantation contributes to the normalization of the state of hipophysal-thyroid systems. Within the early terms, the influence mainly on the extrathyroid system of T3 metabolism is revealed, the direct influence on formation of T4 was revealed later on. At the same time, the final renewal of thyroid structure to the state characteristic of histological structure of intact animal's thyroid did not take place. In rats with hypothyroidism, the natural and synthetic compounds observed and the substances of biological origin produce stimulating effect on the thyroid gland. The introduction of placenta preparations in different forms both in injections and transplantations, contributes to hormonal activity evaluation and thyroid morphological state normalization. On early stages more significant changes were marked in the group of animals, which were administered the injections of placenta, what indicated to higher bioavailability of this form. On further stages the effect of placenta transplantation was saved longer. The stimulating effect of YAV-99 on thyroid activity can be compared to the action of biological preparations. The methods of synthesis of -aryl-3,4-dihydro-4- (2,4-dioxo-6-methylpyran-3-ylidene)- 1,5-benzodiazepines, 5-benzyl-3-methylthiazoline-2-thiones, 5,6-disubstituted7-(3-hydroxiphenyl)-4,7-dihydrotriazolo [1,5-a] pyrimidines and isomer dihydrotriazolo[4,3-a] pyrimidines were worked out. In the latest test researches on intact rats new chemical compound YAV-991 with a thyroid stimulating affect was discovered.
