The chicken domain of alpha-globin genes consits of three alpha-globin genes surrounded by eight housekeeping genes. We found that in contrast to housekeeping genes ggPRX, MPRL28 and AXIN1, expression level of TMEM8 gene is higher in proliferating erythroblasts HD3 than in chicken embryonic fibroblasts and DT40 lymphoid cell line. The steady state level of mRNA of alpha-globin gene HBA1 is 10 times higher than the steady-state level of TMEM8 mRNA. In contrast to human TMEM8, the transcription of chicken TMEM8 is induced approximately five times upon differentiation of HD3 cells. However, the levels of MPRL28 and AXIN1 expression are not changed significantly upon differentiation. The structure of chicken TMEM8 gene and splicing of its pre-mRNA were also analyzed. Erythroid-specific regulatory elements were identified in the body of chicken TMEM8 gene.
