It was shown that overexpression of adaptor protein Ruk/CIN85 lead to increase of HIF-1? intracellular content and its ability to stimulate the luciferase gene reporter activity with HRE elements of erythropoietin gene promoter. Overexpression of Ruk/CIN85 is also accompanied by increase of intracellular content of PAI-1 protein under normoxia and more substantially under hypoxia. The stimulation of PAI-1 expression under Ruk/CIN85 overexpression depends on the presence of HRE site in PAI-1 promotor. Both increase of HIF-1? content and enhancement of PAI-1 expression do not observe after suppression of high Ruk/CIN85 expression level using RNA-interference approach.
