Aim of the investigations: to develop culture protocol for expansion of the hematopoietic stem cells and investigate their morphological and functional characteristics of the early hematopoietic progenitor cells and their immediate progeny during the process of their proliferation and differentiation; to develop mathematical model of proliferation of the self-renewed systems of the organism. Methods: hematopoietic and stromal cell culture in vitro and in vivo, clonogenic analysis in semisolid agar conditions for determination of the functional activity of the hematopoietic stem cells in culture, light microscopy, statistical methods. Theoretical and practical results: a new way of cultivation of hematopoietic stem cell progenitors is developed. Morphological and functional state of the stem cells as well as proliferative potential were determined, and scientific background knowledge was gained as for their use as an alternative source of the stem cells for allogenic transplantation for the patients with various pathological states. Mathematical proliferation model of the self-renewed systems was developed. Novelty: possibility of objective evaluation as the potential transplant of stem cells and progenitors, and as a result, selection of the most appropriate transplantation material, which is most capable of effective hematopoiesis reparation. Efficacy of implementation: achieved scientific data can be used as a background for development of the expansion protocols for hematopoietic stem cells in vitro culture to obtain sufficient quantity for further uses in experimental and clinical research.
