Purpose of work - to identify and estimate by a set of genetic markers the genetic diversity of Trematoda family Opisthorchiidae - Opisthorchis felineus, Metorchis bilis, Pseudamphistomum truncatum, Metorchis xantosomus and to provide a basis for a multilateral and complex research on gene systematics, molecular parasitology, molecular epidemiology of liver flukes. Research Methods - parasitological, biological, microscopic, molecular genetic, epizootological. As a result of research collecting amples of liver flukes of Opisthorchidae family in Ukraine in the basins of the Dnieper, Seversky Donets, Southern Buh, and Danube has been carried out. As a result, there has been found that four kinds of opistorhida circulate in Ukraine - Opisthorchis felineus, Pseudamphismomum truncatum, Metorchis bilis, Metorchis xanthosomus. A collection of Opistorchida DNA samples has been created. The final volumes of DNA extracts were from 50 to 100 ?L and the concentration of total DNA was 120-180 micrograms / ml. After determination of species affiliation of DNA fthere was was carried out amplification of fragments of variable interspace sites ITS1 and ITS2 Opisthorchis felineus, Pseudamphismomum truncatum, Metorchis bilis and Metorchis xanthosomus. Obtained amplicons after electrophoresis were extracted from agarose gel and re-precipitated by adding two volumes of 95% ethanol for sequencing.
