In all cases we haven't observed any significantly increased resistance of these cell lines towards landomycin E compared to wild type parental cells. It is shown that landomycin Е possesses principally different mechanism of anticancer action towards tumor cells in comparison to structurally similar drug doxorubicin, which is already used in clinics. In particular, landomycin E blocks tumor cells in G1-phase of cell cycle, and this process is mediated via reactive oxygen species. The key target of landomycin E is effector apoptotic enzyme caspase-7. This unique mechanism of action allow landomycin E to effectively kill tumor cells, even if they are deficient by key apoptotic enzymes, such as caspase-3. Thus, taking into consideration significant antineoplastic potential of landomycin E and its unique mechanism of action, this antibiotic can be used as second line defense drug during treatment of cancer patients.
