The purpose of work - development of rapid diagnostic and screening of tolerant and resistant to the virus sharka cultivars of peach, apricot, cherry plum and plum at the South and Southeast of Ukraine to be included the tolerant genotypes to in vitro breeding, to creation of immune forms and to propagation of virus-free clones. Methods of investigation - plants-indicator method, ELISA-test, immunoassay analysis, thin layer chromatography (quantitative and qualitative analysis of steroidal glycosides), the tissue and organ culture, in vitro breeding. Results and novelty - the screening and monitoring gave possibility to identify four centers of the virus sharka affection of stone fruits cultivars and hybrids at the South and Southeast of Ukraine, and to assess the degree of affection of gene pool collection and industrial plantations of peach, apricot, cherry plum and plum. PPV tolerant 4 cultivars of apricot, 4 cultivars of peaches, 3 cultivars of cherry plums and 3 cultivars of plums have been selected. Methodological guidelines of tolerant cultivars of peach, apricot, cherry plum and plum to the virus sharka selection. Twelve nutrient medium for obtaining and propagation of immune forms and virus-free cultivars of stone fruits has been modified. On the base of used and improved complex methods for rapid diagnostic, testing and selection of tolerant cultivars the express diagnostic of virus sharka stone fruits system has been developed.
