There have been synthesized new acyl-derivatives of beta-cyclodextrin (CD): succinyl-, maleyl- and aconityl-beta-CD. Intermadiate crystal products of the reaction of succinylation have been studied by X-ray analysis. There have been studied the peculiarities of the reaction of succinylation. There have been studied the antimicrobial activity for synthesized acyl derivatives of beta-CD. For succinyl-beta-CD, appreciable activity has been detected both against gram-positive (S. aureus ATCC 25923) and gram-negative bacteria (E. coli ATCC 25922 та P. aeruginosa ATCC 27853) as well as weak activity against fungi (C. albicans 885-653). Dependence of antimicrobial activity on degree of substitution has not been detected. Molecular modeling of non bonded supramolecular complexes of succinyl-beta-CD and beta-CD has been carried out to study possible molecular mechanisms of antimicrobial activity of cyclodextrins. For succinyl-beta-CD and beta-CD molecules, there has been shown a possibility to self-associate into submolecular aggregates, supramolecular "sponges", which may be formed on the microbe cell surface, absorbing substances supplied to the membranes and leading to disturbance of trans-membrane transport and cell viability. For succinyl-beta-CD supramolecules, there has been shown the possibility to penetrate into the cavity of trans-membrane channels of bacterial membranes, form non bonded complexes with corresponding porin peptides and, as a consequence, block these channels as a possible mechanism of cyclodextrins antimicrobial activity.
