It was established i) that 3 out of 71 synthesized compounds demonstrated clearly expressed dissociation between relaxation of VSM in corresponding intracellular calcium concentration trasnsient; ii) that human mesenchymal stem cells hMSC demonstrate a clearly expressed ability to normalize vascular function damaged following irradiation, i.e. to decrease an elevated arterial blood pressure and myofilament Ca2+ sensitivity, and to repair BKCa function and endothelium-dependent relaxation in vascular tissues obtained from irradiated animals. Thus, hMSC in prospect appear to be worthwhile therapeutic approach in case of ionizing irradiation accident or radiation beam therapy; iii) PKC-?-isoform gene deletion using RNA interference effectively normalized vascular function damaged in arterial hypertension. To the best of our knowledge this is first experience that siRNA-induced PKC-?-isoform gene silencing appears to be a good therapeutic approach to suppress PKC gene encoding function, to inhibit abnormally increased PKC activity, and this way to restore vascular contractility in hypertensive rats.
