To make an immunization of mice with obtained antigens Methods - biochemical, molecular, genetic, genetic engineering. cDNA of potential autoantiges that were identified by SEREX-analysis of antigenic repertoire of medullary breast carcinoma (RAD50, TOP2B, FAM50A, LGal3BP, HMGN2, PARD3, SAP30BP, ANKRD11, SPP1, PDCL, PABCP4, RBPJ, LRRFIP1) were cloned in vectors for expression in bacterial cells (pGEX4T3, pET42b, pET28b). Purified recombinant proteins and fragments of potential autoantigens associated with medullary breast cancer were obtained: С/RAD50, RAD50(36), TOP2B, FAM50A, LGal3BP, HMGN2, PARD3, SAP30BP, ANKRD11, SPP1, PDCL, PABCP4, RBPJ, LRRFIP1, NY-BR-1, NY-BR-62, NY-CO-58. Also purified recombinant protein NaPi2b potentially associated with thyroid cancer was obtained. Mice were immunized with recombinant proteins С/RAD50, FAM50A, LGal3BP, PABCP4, RBPJ, LRRFIP1 and NaPi2b for obtaining of polyclonal antibodies.
