Karazin popup.description2 1-, 3-, 24-, 28-, 30-32- and 35-44-month-old Wistar male rats and isolated hepatocytes of the 3-month-old animals treated by ceramides or thyroxine or melipramine or melipramine and thyroxine as well as blood serum, skeletal muscle, diaphragm, brain cortex, hippocampus, hipotalamus, reticular formation of un-treated or calorie-restricted animals or emotional stress-treated rats were used. Spectrophotometric, radioisotope, electro-physiological, chromatographic methods and "open field" test and shuttle box were used in the work. The aim of the work was to study the key pathway of sphingolipid and phosphoinositide turnover role in the regulation of thyroxine and insulin signal transduction and the calorie restriction effect on sphigolipid metabolism in the brain and behavioral and cognitive functions of old rats and bioelectrical activity of brain structures of rats with low level of movement activity under emotional stress. It has been determined that increased level of ceramide in studied tissues was associated with the organism aging. Ceramide accumulation in the old cells reduced thyroxine and insulin phosphoinositol 3-kinase-dependent signal transduction. The calorie restriction modulates the age-dependent changes of sphingolipid metabolism and cognitive function of the animals. Chronic treatment of rats by calorie restricted diet decreased ceramide content in the hippocampus, facilitated the learning ability of young rats and had no effect on long-term memory of old animals. The calorie restriction reduced negative emotionality and increased investigative activity of old rats as compared with young ones.
