The aim of the work : isolation of highly purified oxidoreductases necessary to create amperometric multi-sensor. The aim of the stage in 2011 year : isolation of the enzymes -the modified forms of alcohol oxydase (AOm) with altered Michaelis constant glycerol oxidase (GO), recombinant flavocytochrome b2 (FC b2), recombinant glycerol dehydrogenase (GDG), as required elements to create biosensitive sensors. Methods of investigations: microbiological, biochemical, physico-chemical, analytical, enzymatic, biosensoric. Obtained results of the work: optimized conditions for GO and GDH producers cultivation mould fungus B. allii 100 (5) and recombinant strain of S. cerevisiae W303 (GDH), respectively; obtained in preparative scale biomass of the producers with a high content of target enzymes for bioanalittycal purpose; isolated and purified preparation of AOm, GO, FC b2 and GDG using previously developed schemes; apprated the possibility of using recombinant GDG H. polymorpha and cells with a damaged cell wall of the strain B. allii as bioselective elements of amperometric sensors for glycerol assay; preparative quantity of highly stabilized enzymes AOm, PO, FC b2 and HDH were transfered to colaborator partner (Laboratory of Biomolecular Electronics Department of Translation Mechanisms of Genetic Information IMBG Ukraine) for testing them as the part of a multibiosensor.
