It was optimized protocol of purification and separation of nanoscale polymer systems for delivery of nucleic acids. It was synthesised polymers based on poly-DMAEM. It was investigated DNA-binding properties of polymers, the optimum parameters of the solution ionic strength, pH, time and temperature regimes for optimal binding with plasmid DNA. It was optimized a protocol of foreign DNA delivery based on the using of poly-DMAEM carriers into plant's protoplasts, in particular, Nicotiana tabacum. It was studied the effect of duration of treatment (time of cultivation of carrier with plasmid DNA), the concentration of polymeric poly-DMAEM carrier and concentration of DNA on the survival of the protoplasts. It was elucidated the cytotoxicity of the developed polymeric carriers during regeneration of protoplasts in culture in vitro.
