Object of the investigations is the cells of the glioma - most hardly curable type of brain tumors. Using of modern methods of expression genetics, Serial Analysis of Gene Expression (SAGE) and microarray analysis, the genes were isolated and characterized, which expressiuon is changed in glial tumors. Obtained results were confirmed by Northern analysis and real time PCR and expression profiles of these genes in astrocytic gliomas were estimated. Rat glioma C6 and human glioma U87MG cell lines were prepared, morphologically characterized and get ready for the evaluation of redox properties of the cells. Influence of marker proteins CHI3L1 and CHI3L2 on the mitogenic properties and proliferative potential of the cells were investigated. It was found that CHI3L1 addition to culture medium stimulates cell division and, in contrast, CHI3L2 addition inhibits cell growth. Recombinant CHI3L1 protein and native CHI3L1 glycoprotein were isolated for the investigation of itheir influence on the redox regulation of the cells. Recombinant cell line 293_CHI3L1, stably producing CHI3L1 protein, was prepared and characterized.
