Aim of the investigations: functional assessment of hematopoietic cells population, which are capable to restore hematopoiesis in case of auto- and allotransplantation. Methods: hematopoietic cell culture in vitro, clonogenic analysis in semisolid agar, morphological methods, variation statistics. Theoretical and practical results: analysis of hematopoietic progenitors colony-forming efficiency in cell culture in vitro was performed, as well as morpho-functional characteristics of the closest progeny of stem cells. The most perspective source of stem cells was cord blood. The most appropriate for using in culture was the combination of interleukin-3, interleukin-6 and granulocyte-macrophage factor, which provided high level of colony-forming activity of cord blood cells in vitro. Novelty: possibility of objective evaluation of functional activity of stem and progenitor cells as the potential transplant. Efficacy of implementation: obtained scientific data can be used as a background for development of the expansion protocols for hematopoietic stem cells in vitro culture to obtain sufficient quantity for further uses in experimental and clinical research.
