During first phase were obtained and purified recombinant proteins tau, RAD50 and NY-BR-1 which will be used for immunization of animals and production of antibodies, essential component of test-kits. cDNA fragments of testicular cancer antigens NY-BR-1 and RAD50 and cDNA fragment of neuron-specific protein tau (MAPT) were subcloned in plasmid vectors for protein expression in prokaryotic cells. Then, fragments of recombinant protein tau (N-terminal part, middle part and C-terminal part) and remombinant proteins C/RAD50(7), RAD50(36), and NY-BR-1 were obtained and purified using affinity chromatography in amounts sufficient for production of monoclonal and polyclonal antibodies. First immunization of animals with above-mentioned recombinant proteins was performed.
